Ayame is the grand-daughter of Dr Genzai, whose surgery Megumi works at. She is good friends with the Kenshin-gumi and thinks of Kenshin as her big brother. She also has a sister called Suzume who also treats Kenshin as a big brother. Overall she treats all the Kenshin-gumi as family because she doesn't have any surviving family except Dr Genzai.
